EGF Overview

Epidermal growth factor (EGF) is a polypeptide composed of 53 amino acids. It is synthesized as a precursor protein with 1217 amino acid residues. It exerts biological activity after hydrolysis and is present in salivary gland and human urine extracts [1]. EGF can induce the premature development of mouse teeth and eyelids, inhibit gastric acid secretion, and promote epidermal growth and keratinization. Although the polypeptide sequences of human EGF (hEGF) and mouse EGF (mEGF) differ at 16 sites, their biological activities are basically the same and they can bind to the same membrane receptor. EGF and its receptor signal are also closely related to the occurrence and prognosis of tumors. EGF receptor inhibitors can be used to treat malignant tumors such as small cell lung cancer and gastric cancer [1-3].

Biological functions of EGF

EGF activates a variety of enzymes after binding to receptors on the cell membrane to exert its biological effects. It has a chemotactic effect on inflammatory cells, fibroblasts, epidermal cells and vascular endothelial cells, and can promote the synthesis of DNA, RNA and hydroxyproline, regulate protein synthesis, improve the construction of collagen, and accelerate the healing of wounds [4]. Studies have found that EGF is a powerful cell division promoting factor that has a wide range of effects on promoting the division and proliferation of epithelial cells. It acts on the wound surface in an autocrine and paracrine manner, promoting the division and proliferation of repair cells such as epidermal cells and fibroblasts, promoting wound re-epithelialization, and also has a certain induction effect on angiogenesis [5]. At the same time, EGF can also stimulate epithelial cells to synthesize and secrete extracellular matrices such as collagen and hyaluronic acid, promote the growth of connective tissue cells, promote the formation of new granulation tissue and wound re-epithelialization, accelerate wound healing of various tissues such as skin, cornea and gastrointestinal tract, and is closely related to liver regeneration and wound repair [6].
